In comparing the specs for my second generation Prius with the third generation, it appears that the cargo space has increased about 35%. With loosing the under deck space, this would be nice. Have those who have gone from a second to third generation Prius notice this significant change, or is it just specsmanship?

No...go to Toyota Canada website to compare specs on same basis. Gen3 gained about 1+ft3 which is not bad.
Around 2010, just when Gen3 was released, the US autos starting reporting a new spec known as "EPA Cargo Capacity". For some reason (in my mind, possibly to help EV's look better) EPA allowed the US autos to report larger cargo capacity. It must be EPA allows one or two rear seats down. If you look at Leaf specs, it has humongous cargo capacity (spec only of course).
