Does connecting my iPhone to my car's cigarette lighter to charge it affect mpg?

In theory YES but in practice NO.

Every energy including what charges your iphone comes from the fuel. iPhone GPS or the radio will have no measurable effect on MPG.

But running light may ... 110W if you running the stock lowbeam but of course you can not save on that

It will but only by about as much as the weight of the iPhone will affect MPG. In other words, too small to measure.
