I just bought a 2006 Prius with one FOB.
How do I know if my 2006 Prius is SKS?
The fob unlocks the doors by pushing the button.
There is no reaction to the FOB being in my pocket in trying to start the car.
Is my FOB non-SKS but my Prius is?

Is there some part of the VIN that says SKS?
    Thanks in advance.

    Hi and welcome to PC.
Your car has SKS if the fob has a silver logo on the back and the car has a button on the door handle. If the logo is black and the door handle has no button you do not have SKS.
Please put a location in your avatar as this affects the vehicle you have and the help we can give.

If you have SKS as described above and it's not working, check to see if it's been turned off at the button under the steering column.

As Bisco says you would be well advised to get another fob as soon as possible. The Prius is programmed to the fob, but to do this you must have a working fob. In this situation you can do this yourself.
Loosing the only fob you have means a tow to the dealer, buying a new fob, the dealer will then charge you a lot of money to re seed the security system to be able to program the car to the new fob.

    Thank you very much.
The button under the steering wheel worked just fine.
