After swapping out 2 engine in prius Gen 3 the ac doesn’t work. If I disconnect neg 12 volt and orange disconnect from battery pack. Let seat for 15 minutes do you think I need 1000 volt gloves to take out fuse and check continuity? Thanks

Before trying that, or trying anything, I would start by asking the HVAC system for any trouble codes. (You haven't said a lot about just what sort of "doesn't work" you are seeing.)

Start with the car off. Hold down both the A/C AUTO and FRESH/RECIRC buttons, while turning the car on (two power button presses, no foot on brake). Look for two-digit codes shown at the left end of the HVAC display, as described near the bottom of this page:

I tried to do that and it didn’t work but I’ll follow your instructions and see if it’ll give me any codes. Thank you

If you've removed the fuse to check continuity, why would current be flowing across it? Now pulling the fuse out, you'll need to be careful of residual current in the system.
